| 生化机理 | Novobiocin Sodium (U-6591) is a small molecule inhibitor of eukaryotic DNA topoisomerase II and prokaryotic DNA gyrase. In the case of DNA gyrase inhibition, Novobiocin functions as a competitive inhibitor of the ATP binding event necessary for turnover of the enzyme after a manipulation of DNA supercoils. Inhibition of these enzymes blocks the mechanism for detangling DNA and establishing the proper super-coiled state necessary for normal replication and transcription, compromising the viability of the cell. Treatment with Novobiocin is shown to potentiate the cytotoxic effects of DNA alkylating agents. Novobiocin is also described to inhibit the activity of retroviral RNA-dependent DNA polymerase. Novobiocin is an inhibitor of Hsp90. Novobiocin interacts with the C-terminal ATP-binding domain of Hsp90.Antibiotic. Potent, competitive DNA gyrase inhibitor (IC 50 = 80 nM). Inhibits organic ion transporters and Hsp90. Active in vitro and in vivo . |
