Aeroplysinin 1 shows anti-proliferative effect against tumor cells (HT-1080, HTC-116, HeLa, THP-1, NOMO-1 and HL-60 cells), with IC₅₀s ranging from 2.3 to 17 μM^[1].
Aeroplysinin-1 also exhibits an antiviral activity toward HIV-1 caused by inhibition of its reverse transcriptase activity^[1].
Aeroplysinin 1 inhibits P. phosphoreum, C. wailesii, P. minimum and HIV with IC₅₀s of 3.5, 5.6, 7.0 and 14.6 μM^[1].
Aeroplysinin 1 inhibits human endothelial cells (EVLC-2, HMEC, RF-24, and HUVEC cells), with IC₅₀s ranging from 2.6 to 4.7 μM^[2].
(+)-Aeroplysinin-1 (0.25-0.5 μM) blocks the EGF-dependent proliferation of both MCF-7 and ZR-75-1 human breast cancer cells and inhibits the ligand-induced endocytosis of the EGF receptor in vitro^[3].

生物活性	Hirsutenone is an active botanical diarylheptanoid present in Alnus species and exhibits many biological activities, including anti-inflammatory, anti-tumor promoting and anti-atopic dermatitis effects. Hirsutenone attenuates adipogenesis by binding directly to PI3K and ERK1 in a non-ATP competitive manner. Hirsutenone can be used for the study of obesity^[1].
体外研究 (In Vitro)	Hirsutenone (0-100 μM; 48 hours) inhibits adipogenesis in 3T3-L1 preadipocytes and exhibits non-toxicity at 20-100 μM^[1]. Hirsutenone (0-100 μM; 48 hours) attenuates MDI-induced lipid accumulation in 3T3-L1 preadipocytes in a dose-dependent manner. In particular, HST at 40 and 80 μM significantly reduces MDI-induced adipogenesis in 3T3-L1 preadipocytes^[1]. Hirsutenone (0-100 μM; 48 hours) reduces the protein expression levels of PPARg, C/EBPa, and FAS in a dose-dependent manner in 3T3-L1 preadipocytes^[1]. Hirsutenone (80 μM; 20-24 hours) suppresses the cell cycle entry to S and G2/M phases occurrs at 20 hours when compares with at 20 hours. At 24 h, more cells are arrested in G1 phase (53% of total cells) when compares with the MDI-induced group which contains the majority of cells (56% of total cells) in G2/M phase^[1]. Description:

Maleimide functionalized polyethylene glycol, Maleimide PEG is a sulfhydryl (thiol/SH) group reactive PEG derivative that can be used to selectively modify protein, peptide or any other surfaces with availablesulfhydryl groups. The reaction between maleimide and thiol proceeds readily at pH 6.5~7.5 to form stable thioether bonds.

Physical Properties:

Off-white/white solid or viscous liquid depends on molecule weight;
Soluble in regular aqeous solution as well as most organic solvents;

Storage Conditions:

Store at -20 ⁰C, dessiccated. Avoid frequent thaw and freeze.

Reaction Procedures:

Generally, a 10- to 20-fold molar excess of Maleimide PEG over the amount of thiol-containing material results in sufficient conjugation.

Materials Required:

Pegylation buffer, PBS buffer, pH 7 or other thiol-free buffer at pH 7.
Maleimide stock solution: 100 mg in 1 mL conjugation buffer.
Washing solution: Distilled water or any aqueous buffer.

Reaction Steps:

Dissolve targeted materials in conjugation buffer. Estimate the concentration thiol groups on the targeted materials. Add maleimide PEG stock solution to the targeted conjugation materials with the final concentration keep at least 10 mg/mL. 10~50 molar excess of PEG maleimide needed for optimal conjugation; Allow mixture agitates at room temperature for 2~4 hours at room temperature or overnight at 4 ⁰C.Conjugate can be purified either by size exclusion chromatography or dialysis.
