一般描述

脱氧野霉素（AMP-dNM）是dNM的疏水衍生物，可有效抑制β-葡萄糖苷酶2（BGD，IC | 50 | = 0.3 nM），对葡萄糖苷神经酰胺合酶的拮抗作用较小（GCS，IC | 50 | = 25 nM），并且几乎不能抑制其他GCase亚型。脂质信使神经酰胺通过GCS转化为葡萄糖基神经酰胺。在相反的方向上，BGD从葡糖神经酰胺中切割出葡糖基部分，释放出神经酰胺，后者可以转化为鞘磷脂。AMP-dNM已显示可诱导Hep-G2细胞中固醇调节元件结合蛋白调节的基因表达和胆固醇合成，在半抗原诱导的结肠炎的鼠模型

AMP-Deoxynojirimycin (AMP-dNM) is a hydrophobic derivative of dNM that potently inhibits β-glucosidase 2 (BGD, IC|50|= 0.3 nM), less potently antagonizes glucosylceramide synthase (GCS, IC|50|= 25 nM), and only poorly inhibits other GCase isoforms. The lipid messenger ceramide is converted to glucosylceramide by GCS. In the reverse direction, BGD cleaves the glucosyl moiety from glucosylceramide, liberating ceramide, which can be converted into sphingomyelin. AMP-dNM has been shown to induce sterol regulatory element-binding protein-regulated gene expression and cholesterol synthesis in Hep-G2 cells, strongly suppress inflammation in a murine model of hapten-induced colitis, and enhance insulin sensitivity in murine and rat models of insulin resistance.

The Temozolomide (TMZ)-resistant cell line LN18 is treated with a combination of TMZ and 4-Diethylaminobenzaldehyde for 10 days. As indicated by colony formation assay, the clonogenic capacity of LN18 cells exposed to TMZ in combination with 4-Diethylaminobenzaldehyde (DEAB) is significantly attenuated[3].

Carvedilol (BM 14190) is a non-selective β/α-1 blocker[1]. Carvedilol inhibits lipid peroxidation in a dose-dependent manner with an IC50 of 5 μM.     Acetylthio PEG NHS, also called thiolacetate PEG succinimidyl NHS ester, is an amine reactive PEG with protected thiol groups. Acetyl protected thiol group is stable in neutral condition and acetyl group can be deprotected from thiol group under acidic or alkaline conditions. Free thiol group is released upon deprotection of acetylthio. With thiol group protected, remaining functional groups, such as amines, carboxylic acids, azides, hydrazides, NHS, maleimides, DBCO, etc. can be used to react with other groups. PEGylated thioacetate derivatives are water soluble and can be used directly in aqueous buffer.

    Product Description:

    Alexa Fluor 488 labeled Proteinase K is one of Nanocs’ fluorescent Proteinase K conjugates. Proteinase K is a highly reactive nonspecific serine protease that is capable of inactivating RNases and DNases and is used in the isolation or preparation of high molecular weight nucleic acids. Proteinase K is also useful for helping to characterize enzymes, due to its cleavage specificity. Proteinase K is stable in a wide variety of detergents and buffer salts and at various temperatures and pH. The isoelectric point of proteinase K is 8.9. Nanocs provides a number of fluorescent labeled Proteinase K products that span from UV region to near IR. These fluorescent Proteinase K conjugates can be used for enzyme studies, bioassay development and many other biomedical research. Nanocs Alexa Fluor 488 labeled Proteinase K is a green fluorescent Proteinase K conjugate that has excitation/emission wavelength at 495/515 nm. This Alexa Fluor 488 Proteinase K can be easily detected by fluorescent instruments in FITC channel. All our fluorescent Proteinase K conjugates were purified by size chromatography to remove non-conjugated molecules and ensure adequate applications both in-vitro and in-vivo.
